The measurement of o-tyrosine from exhaled breath condensate (EBC) is a non-invasive method for detecting amino acid oxidation associated with silica- and asbestos-induced diseases. O-tyrosine is an amino acid oxidation marker; it has not been described extensively. A method was developed for the selective and sensitive detection of o-tyrosine using liquid chromatography and electrospray ionization-tandem mass spectrometry. The LC-ESI-MS/MS detection method described here is highly specific for quantifying o-tyrosine in clinical samples from asbestosis/silicosis patients. This method consists of a pre-treatment procedure and lyophilization, intended for concentrating biomarkers from EBC. Enzymatic reactions, temperature fluctuations, and oxidation are all common reasons for degradation of clinical samples. The addition of antioxidant, rapid sample preparation under mild conditions, and storage of sample at −80 ◦C in sealed sample vials could all prolong the storage period of samples without marked deterioration of o-tyrosine biomarker for up to a month. The developed LC-ESI-MS/MS method was used to assess the extent of oxidative stress, by determining o-tyrosine concentration, in EBC of subjects with long-term exposure to asbestos or silica dust, compared to control subjects. These subjects were of similar average age and the same gender. The concentration levels of o-tyrosine were statistically different between asbestosis and silicosis patients compared to healthy control subjects. ( J Breath Res.
