Description of 12-HETE with associated disease summary

12-hydroxyeicosatetraenoic acid (12-HETE) is one of specialized proresolving mediators (SPMs), which play a critical role in the active resolution of inflammation without immunosuppression. Endogenous 12-HETE is produced by enzymatic oxygenation of polyunsaturated fatty acids. It is known to participate in a chemotaxis of neutrophils in the inflammatory process. Level of 12-HETE is reported to be dysregulated in patients with chronic obstructive pulmonary disease (COPD). 12-HETE level decreases in EBC, but increases in bronchoalveolar lavage fluid (BALF) (Am J Physiol Lung Cell Mol Physiol. (2015)). Level of EBC 12-HETE increase in asthma subjects. However, no significance difference in the level of EBC 12-HETE is observed between asthma and aspirin-induced asthma subjects (J Allergy Clin Immunol. (2011)). Churg-Strauss syndrome (CSS) can be clearly distinguished from healthy controls, bronchial asthma, and hypereosinophilic syndrome (HES) by a significant increase in EBC 12-HETE concentration. Clinical activity of CSS does not correlate to 12-HETE levels in EBC (Clin Exp Allergy. (2012)). Most of the studies use high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) to identify EBC 12-HETE. Gas chromatography-mass spectrometry (GC-MS) and lipid-mediator metabolipidomics followed by MS/MS are also used in detection of EBC 12-HETE. The non-invasive detection of 12-HETE level in EBC may monitor anti-inflammatory intervention and the progress of asthma. EBC 12-HETE is proved to be a highly efficient biomarker differentiating CSS and shows a potential to be utilized in diagnosis of CCS in the early-stage. Asthma

Concentration of EBC 12-HETE increases in asthma subjects.

Aspirin-induced asthma

EBC 12-HETE increases in aspirin-induced asthma subjects. There is no significance difference in the level of 12-HETE between asthma and aspirin-induced asthma subjects.

Chronic obstructive pulmonary disease (COPD)

EBC 12-HETE level decreases in COPD subjects. However, 12-HETE level is reported to be dysregulated in COPD subjects.

Churg-Strauss syndrome (CSS)

A significant elevation of 12-HETE EBC is observed in CSS subjects in contrast to healthy controls, asthma and HES. The increase in 12-HETE is independent of the activity of CSS.
